Quick answer
In two trials, blood NAD+ was higher after 14 days of NMN, and in a trial of 250 mg a day it was higher at the first check, after 4 weeks. Breakdown products show up in the blood within hours of a dose. Effects you can feel, or health changes beyond blood NAD+, have not been shown.1,2,3,4
Key facts
- Four weeks after the last dose, blood NAD+ was back where it started.3
- The longest NMN trials ran 24 weeks.5,6
- How much NAD+ rose varied widely between people on the same amount.7
What changes, and when
| Time after starting | What was found | Who and how much |
|---|---|---|
| Within 5 hours of one dose | Breakdown products of nicotinamide rose, more at higher doses | 10 healthy men, 100 to 500 mg |
| 8 and 14 days | Compounds made from NAD+ higher than at the start | 32 adults aged 55 to 80, 1,000 mg once or twice a day |
| 14 days | NAD+ up by similar amounts with NMN or nicotinamide riboside | 65 healthy adults |
| 4, 8 and 12 weeks | NAD+ higher at every check | 30 adults, 250 mg a day |
| 30 and 60 days | NAD+ higher in every dose group | 80 adults, 300 to 900 mg a day |
Blood NAD+ was higher after 14 days of NMN in two trials, one of them giving 1,000 mg once or twice a day.1,2
In a 12-week trial of 250 mg a day, blood NAD+ was already higher at the first check, after 4 weeks, and stayed higher at 8 and 12 weeks.3
How long it stays in your system
Four weeks after the last dose, blood NAD+ was back at its starting level.3
So the higher level lasts only while you keep taking NMN. The trial did not check how fast NAD+ fell in the days after the last dose, only that it was back to normal four weeks later.3
The vitamin B3 part of NMN ends up as the same breakdown products as niacin, and these leave the body in urine.5
When would you notice anything?
Raises NAD+ levels in the blood*5,8,9
Moderate evidence
A meta-analysis of 12 randomized trials (513 adults) found that NMN raised blood NAD+, and a 60-day trial in 80 healthy adults aged 40 to 65 found increases at 300, 600 and 900 mg a day. The trials were small and short, several were funded by NMN makers, all carried some risk of bias, and the same meta-analysis found that most health outcomes were no different from placebo.
Amount studied: 250 to 900 mg a day, for up to 12 weeks
Blood NAD+ is a lab value, so there is nothing to feel when it rises. Trials that tested function, such as walking tests or grip strength, ran 6 to 12 weeks and gave mixed results.8,10,11
In trials of adults over 60, NMN did not change grip strength, walking speed or muscle mass; nearly all of these trials used 250 mg a day.11
Why results vary from person to person
Among people taking the same amount in a 60-day trial, the rise in blood NAD+ varied widely from person to person.7
Trials also measured different things, in whole blood, plasma, serum or blood cells. NAD+ and related compounds went up in all of them, while NMN itself went up in some and not in others.5
What’s in ours
- NMN: β-Nicotinamide Mononucleotide, 500 mg per serving, from the label
Starting and stopping
Use for longer than 24 weeks has not been studied in trials, so long-term effects are unknown.5,6
- If you take medicine for diabetes, ask your doctor before starting: insulin after lunch rose over 12 weeks of daily NMN in a small study12
- If you are pregnant, breastfeeding or under 18, ask your doctor first
Questions people ask
How long does NMN stay in your system?
Blood NAD+ stayed higher while people took NMN every day and was back at its starting level four weeks after they stopped. Earlier points after stopping were not measured.3
