Quick answer
Most NMN trials used 250 mg a day, and trials have tested 250 to 2,000 mg a day, the highest amounts for 6 weeks at most. Europe's food safety agency judged up to 300 mg a day safe for adults. In the one trial comparing amounts, blood NAD+ was highest in the 600 and 900 mg groups.1,2,3,4,5,6
Key facts
- The human safety data Europe reviewed reached 900 mg a day for 60 days, and 250 mg a day for 24 weeks.1
- People taking the same amount showed very different rises in blood NAD+.7
- Counted as vitamin B3, 500 mg of NMN supplies about 183 mg of nicotinamide.1
- Trials gave the daily amount in one dose or as two half doses.6,8
The amounts used in trials
250 mg a day is the amount most NMN trials have used, including nearly all trials in adults over 60.1,9
Randomized trials have given 250 to 2,000 mg of NMN a day, for 14 days to 24 weeks.2
| Who took part | Daily amount | How long |
|---|---|---|
| 30 healthy adults aged 20 to 65 | 250 mg, as 125 mg twice | 12 weeks |
| 36 healthy adults aged 40 to 59 | 250 mg, as 125 mg twice | 12 weeks |
| 42 healthy men over 65 | 250 mg | 6 or 12 weeks |
| 80 healthy adults aged 40 to 65 | 300, 600 or 900 mg | 60 days |
| 48 amateur runners | 300, 600 or 1,200 mg | 6 weeks |
| 31 healthy adults aged 20 to 65 | 1,250 mg | 4 weeks |
| 32 adults aged 55 to 80 | 1,000 mg once or twice | 14 days |
Does more NMN do more?
Raises NAD+ levels in the blood*1,6,12
Moderate evidence
A meta-analysis of 12 randomized trials (513 adults) found that NMN raised blood NAD+, and a 60-day trial in 80 healthy adults aged 40 to 65 found increases at 300, 600 and 900 mg a day. The trials were small and short, several were funded by NMN makers, all carried some risk of bias, and the same meta-analysis found that most health outcomes were no different from placebo.
Amount studied: 250 to 900 mg a day, for up to 12 weeks
In a 60-day trial that compared 300, 600 and 900 mg a day, blood NAD+ rose in all three groups and was highest at 600 and 900 mg.6
Among people taking the same amount in a 60-day trial, the rise in blood NAD+ varied widely from person to person.7
Outside blood NAD+, the evidence on amounts is thin. In amateur runners, breathing-threshold measures changed more than with placebo at 600 and 1,200 mg a day, while maximal oxygen uptake did not change at any amount.3
In adults over 60, nearly every trial used 250 mg, so whether a larger amount would change strength or walking speed in that age group is untested.9
How NMN counts toward vitamin B3
300 mg of NMN supplies about 110 mg of nicotinamide, a form of vitamin B3. That is roughly five times the adult reference intake for niacin, and below Europe's upper level of 900 mg of nicotinamide a day.1
By the same ratio, 500 mg of NMN supplies about 183 mg of nicotinamide and 1,000 mg about 366 mg, our arithmetic from Europe's figure. Both stay below the 900 mg upper level for nicotinamide, but niacin, nicotinamide or nicotinamide riboside from other products adds to the total.1
For scale, adults need roughly 11 to 22 mg of niacin a day, depending on how much energy they use, so even 250 mg of NMN supplies several times that.1
What's in ours
What’s in ours
- NMN: β-Nicotinamide Mononucleotide, 500 mg per serving, from the label
One capsule of ours gives 500 mg. That is twice the most common trial amount, within the range that raised blood NAD+, and above the 300 mg a day Europe assessed.1,12
Before taking more
Europe's food safety agency judged NMN safe at up to 300 mg a day for adults, not including women who are pregnant or breastfeeding.1
At 1,250 mg a day for 4 weeks, healthy adults showed no changes in blood, urine or body measurements beyond normal variation, and no severe adverse events.4
- Ask your doctor first if you are pregnant or breastfeeding, or under 18
- Ask first if you take prescription medicines, especially for diabetes
- Add up the vitamin B3 in any multivitamin or other NAD+ product you already take1
Questions people ask
Is 1,000 mg of NMN a day too much?
It has only been tested briefly: 1,000 mg once or twice a day for 14 days, 1,200 mg for 6 weeks and 1,250 mg for 4 weeks, without serious problems reported. Europe judged up to 300 mg a day safe, and longer use of high amounts has not been studied.1,3,4,5
Should the amount depend on age?
No trial has set amounts by age. Most trials in adults over 60 used 250 mg a day, and the trial in adults aged 40 to 65 used 300 to 900 mg.6,9
Is the amount different for men?
The trials used the same amounts for both sexes. In a 14-day trial, how much NAD+ rose did not depend on sex.5
